Rat Monoclonal Antibody

The monoclonal antibody 11A10-B7 recognizes mouse CD301b. CD301b is also known as macrophage galactose-type C-type lectin 2 (MGL2). CD301b expression is restricted to dermal dendritic cells, connection tissue, and bone marrow derived dendritic cells. Macrophages also express CD301b at low levels. CD301b contains a single carbohydrate recognition domain that recognizes protein antigens that have a galactose and N-acetylgalactosamine (Gal/GalNAc). Gal/GalNAc protein antigens are efficiently internalized through CD301b and presented to antigen specific CD4+ cells via MHC Class II. CD301b+ dendritic cells are believed to play an important role in TH2 priming. Applications Tested: This 11A10-B7 antibody has been tested by immunohistochemistry of frozen mouse tissue and can be used at less than or equal to 0.25 μg/mL. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.Staining Protocol: All protocols work well for this monoclonal antibody. Use of Protocol A: Two-step protocol: intracellular (cytoplasmic) proteins allows for the greatest flexibility for detection of surface and intracellular (cytoplasmic) proteins. Use of Protocol B: One-step protocol: intracellular (nuclear) proteins is recommended for staining of transcription factors in conjunction with surface and phosphorylated intracellular (cytoplasmic) proteins.
