Mouse Monoclonal Antibody

The VU-1D9 antibody specifically reacts with human Ep-CAM, a 40kD epithelial protein expressed on baso-lateral cell surface in many epithelial tissues (but absent from mesothelial tissues). EpCAM-1 is a homophilic and Ca2+ independent adhesion molecule that is expressed on the baso-lateral cell surface of many epithelial, but not mesothelial tissues. It is expressed de novo in squamous cell carcinoma but is absent in the majority of healthy squamous epithelia. EpCAM expression correlates with cell proliferation, dedifferentiation, and the progression of tumorigenicity. VU-1D9 antibody differentiates between mesothelioma and carcinoma, a common diagnostic problem. Applications Reported: Flow Cytometry, Functional Studies (Blocking), Immunohistochemistry (Frozen, Paraffin), Immunoprecipitation, Western Blotting. Applications Tested: This VU-1D9 antibody has been tested by immunohistochemistry on formalin-fixed paraffin embedded human tissue using low pH antigen retrieval and can be used at 5 g/mL. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest. Using conjugate solutions: Centrifuge the protein conjugate solution briefly in a microcentrifuge before use; add only the supernatant to the experiment. This step will help eliminate any protein aggregates that may have formed during storage, thereby reducing nonspecific background staining.

Ep-CAM (epithelial adhesion molecule, epithelial specific antigen, ESA) is a transmembrane glycoprotein expressed in the epithelium with a molecular weight of approximately 40 kDa, which functions as an epithelial cell adhesion molecule. Ep-CAM functions as a homotypic calcium-independent cell adhesion molecule, and has a direct impact on cell cycle, proliferation and metabolism of epithelial cells and fibroblasts due to its ability to rapidly induce the proto-oncogene c-myc and the cell cycle regulating genes cyclin A and E. Ep-CAM mediates Ca2+-independent homotypic interactions. Formation of Ep-CAM-mediated adhesions have a negative regulatory effect on adhesions mediated by classic cadherins, which may have strong effects on the differentiation and growth of epithelial cells. Ep-CAM overexpression was suggested to be associated with enhanced epithelial proliferation. Ep-CAM is highly expressed in human carcinomas, and is a marker for tumors of epithelial lineage. Ep-CAM is expressed on baso-lateral cell surface in most simple epithelia and many carcinoma types. Also, Ep-CAM reportedly distinguishes adenocarcinomas from pleural mesotheliomas.
