DNase I (RNase-free) is a recombinant endonuclease expressed in E coli catalyzing the hydrolysis of single- and double-stranded DNA without sequence specificity yielding oligonucleotides carrying 5 -phosphate and 3 -hydroxyl termini Its enzymatic activity strictly depends on Ca ions while Mg or Mn ions additionally modulate enzymatic preference Mg facilitates random cleavage at any site of double-stranded DNA whereas Mn induces relatively aligned double-stranded cleavage patterns Complete heat inactivation of DNase I occurs at 65 C for 10 minutes in the presence of metal-ion chelators This RNase-free nuclease is commonly employed in RNA isolation procedures in vitro transcription assays and preparation steps prior to RT-PCR to eliminate DNA contamination The product is supplied as a purified DNase I solution free of detectable RNase activity
