Ribonuclease I (RNase I), an endoribonuclease, preferentially hydrolyzes single-stranded RNA to nucleoside 3'-monophosphates via nucleoside 2', 3'-cyclic monophosphate intermediates.

The enzyme does not require any metal ions for activity.
