Filipin III is a naturally occurring fluorescent polyene macrolide antibiotic isolated as the primary active isomer from cultures of Streptomyces filipinensis It binds specifically to free (unesterified) cholesterol in biological membranes through reversible interaction causing alteration of its spectral properties (excitation/emission maxima around 360 nm/480 nm) Due to this cholesterol-binding affinity and inherent fluorescence Filipin III has been extensively employed as a probe for visualizing and quantifying cholesterol distribution and trafficking in cell membranes In biomedical research Filipin III is applied in fluorescence microscopy and flow cytometry to detect free cholesterol dynamics lipid raft formation and lipoprotein metabolism alterations Currently no definitive IC50 values have been reported for Filipin III in typical cellular assay conditions
