Rabbit Polyclonal Antibody

AA Sequence of recombinant protein: MSPDKQMAVL PRRERNRQAA AANPENSRGK GRRGQRGKNR GCVLTAIHLN VTDLGLGYET KEELIFRYCS GSCDAAETTY DKILKNLSRN RRLVSDKVGQ ACCRPIAFDD DLSFLDDNLV YHILRKHSAK RCGCI. Preparation: Produced from sera of rabbits immunized with highly pure Recombinant Human GDNF. Anti-Human GDNF-specific antibody was purified by affinity chromatography and then biotinylated. Sandwich ELISA: To detect hGDNF by sandwich ELISA (using 100 μL/well antibody solution) a concentration of 0.25-1.0 μg/mL of this antibody is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Human GDNF (500-P81) as a capture antibody, allows the detection of at least 0.2-0.4 ng/well of Recombinant hGDNF. Western Blot: To detect hGDNF by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 mg/mL. Used in conjunction with compatible secondary reagents the detection limit for Recombinant hGDNF is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions. 500-P81BT-1MG will be provided as 2 x 500 μg

GDNF is a glycosylated, disulfide-bonded homodimer molecule. It was first discovered as a potent survival factor for midbrain dopaminergic neurons and was then shown to rescue these neurons in animal models of Parkinson's disease. GDNF is about 100 times more efficient survival factor for spinal motor neurons than the neurotrophins. GDNF is a neurotrophic factor that enhances survival and morphological differentiation of dopaminergic neurons and increases their high-affinity dopamine uptake.
