Mouse Monoclonal Antibody

Description: This SL-12 monoclonal antibody reacts with human Interferon Regulatory Factor (IRF)-3. IRF-3 is a 427 amino acid constitutively expressed member of the IRF transcription factor family that plays a critical role in the induction of type I interferons. Inactive IRF-3 continuously transits between the cytoplasm and nucleus. Virus or dsRNA induces the C-terminal phosphorylation of IRF-3 by IKKε and TBK-1. This phosphorylation results in retention of IRF-3 in the nucleus and subsequent transcription of IFN-β, several interferon stimulated genes (ISGs), and other antiviral response genes. Applications Reported: This SL-12 (SL12) antibody has been reported for use in immunoblotting (WB). Applications Tested: This SL-12 (SL12) antibody has been tested by immunoblot of normal human peripheral blood cell lysates. This antibody can be used at less than or equal to 2 μg/mL in blocking buffer. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest. Purity: Greater than 90%, as determined by SDS-PAGE. Aggregation: Less than 10%, as determined by HPLC. Filtration: 0.2 μm post-manufacturing filtered.

IFR3 (interferon regulatory factor 3) is a key transcriptional regulator of type I interferon (IFN)-dependent immune responses. It plays a critical role in the innate immune response against DNA and RNA viruses. IFR3 regulates the transcription of type I IFN genes (IFN-alpha and IFN-beta) and IFN-stimulated genes (ISG) by binding to an interferon-stimulated response element (ISRE) in their promoters. It acts as a more potent activator of the IFN-beta gene than the IFN-alpha gene and plays a critical role in both the early and late phases of the IFNA/B gene induction. IFR3 is found in an inactive form in the cytoplasm of uninfected cells and following viral infection, double-stranded RNA, or toll-like receptor signaling. It is phosphorylated by IKBKE and TBK1 kinases. This induces a conformational change, leading to its dimerization and nuclear localization and association with CREB binding protein to form dsRNA-activated factor 1 (DRAF1), a complex which activates the transcription of the type I IFN and ISG genes.
