A medium for susceptibility testing of methicillin resistant staphylococcus aureus (MRSA)

  • Mueller Hinton II Agar + 2% NaCl is specifically used for testing the antibiotic susceptibility of Staphylococcus spp., particularly for detecting methicillin resistance (MRSA/MRSE) via disc diffusion or gradient test, because the high salt concentration (2% NaCl) promotes the expression of resistance genes (mecA), giving accurate results for these salt-tolerant bacteria.
  • It is standardized by CLSI/EUCAST for this purpose, ensuring reliable detection of resistance phenotypes like methicillin-resistant S. aureus (MRSA). 2% NaCl creates a hypertonic environment, forcing Staphylococcus to express the resistance mechanism, leading to smaller inhibition zones and correctly identifying resistant isolates.
  • This medium is manufactured to contain low levels of thymine and thymidine and controlled levels of calcium and magnesium ions.
