VPC 23019 (CAS 449173-19-7) is a small molecule antagonist targeting sphingosine-1-phosphate receptors S1P1 and S1P3 It disrupts S1P-mediated signaling with an IC50 of 8 nM for S1P1 inhibition VPC 23019 abolishes S1P-induced cellular migration Gi-dependent Rac activation and tube formation in relevant cell models In vascular endothelial cells it does not alter basal nitric oxide production but attenuates Angiotensin II-stimulated NO release The compound also suppresses S1P-induced transendothelial migration and partially inhibits S1P-evoked calcium elevation in fibroblasts In vascular reactivity assays VPC 23019 modulates S1P-induced contraction in endothelium-intact vessels without affecting responses to other vasoactive agents VPC 23019 supports research into S1P receptor functions in cellular signaling and vascular physiology
