Mouse Monoclonal Antibody

Description: The eB114 antibody reacts with the RASSF1A isoform of the tumor suppressor gene RASSF1. RASSF1A contains a Ras Association Domain (RA) and binds Ras in a GTP-dependent manner to effect apoptosis. The RASSF1A isoform undergoes epigenetic inactivation in a variety of human primary tumors. This has been associated with hypermethylation of a CpG island(s) in its promoter region. This antibody was generated against a synthetic peptide corresponding to the splice junction region of RASSF1A. Applications Reported: The eB114 antibody has been reported for use in immunoprecipitation, and immunoblotting (WB). Applications Tested: The eB114-10H1 has been tested by Western blotting and can be used at approximately 1-10 μg/mL. RASS1A is detected as a 45 kDa band in Western blotting of a HeLa cell extract. It is recommended that the reagent be carefully titrated for optimal performance in the assay of interest. Purity: Greater than 90%, as determined by SDS-PAGE. Aggregation: Less than 10%, as determined by HPLC. Filtration: 0.2 μm post-manufacturing filtered.

This gene encodes a protein similar to the RAS effector proteins. Loss or altered expression of this gene has been associated with the pathogenesis of a variety of cancers, which suggests the tumor suppressor function of this gene. The inactivation of this gene was found to be correlated with the hypermethylation of its CpG-island promoter region. The encoded protein was found to interact with DNA repair protein XPA. The protein was also shown to inhibit the accumulation of cyclin D1, and thus induce cell cycle arrest. Seven alternatively spliced transcript variants of this gene encoding distinct isoforms have been reported.
