Rat Isotype Control

Description: The monoclonal rat IgG2a, kappa is useful as an isotype control immunoglobulin. Applications Reported: APC-eFluor™ 780 Rat IgG2a Isotype Control (APC-Alexa Fluor™ 750 replacement) has been reported for use in flow cytometric analysis. Applications Tested: Rat IgG2a K Isotype Control APC-eFluor™ 780 has been tested by flow cytometric analysis of mouse splenocytes. Use isotype control at the same concentration as experimental antibody. APC-eFluor 780 emits at 780 nm and is excited with the Red laser (633 nm). Please make sure that your instrument is capable of detecting this fluorochome. Light sensitivity: This tandem is sensitive to photo-induced oxidation. Please protect this vial and stained samples from light. Fixation: Samples can be stored in IC Fixation Buffer (cat. 00-8222) (100 μL cell sample + 100 μL IC Fixation Buffer) or 1-step Fix/Lyse Solution (cat. 00-5333) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation. Some generalizations regarding fluorophore performance after fixation can be made, but clone specific performance should be determined empirically. Excitation: 633-647 nm; Emission: 780 nm; Laser: Red Laser. Filtration: 0.2 μm post-manufacturing filtered.

Rat IgG2a is an immunoglobulin subclass that is commonly used in research and diagnostic applications. It is produced by the rat immune system and is known for its ability to activate complement and mediate antibody-dependent cell-mediated cytotoxicity (ADCC). Rat IgG2a has a molecular weight of approximately 160 kDa and is composed of two heavy chains and two light chains. It is often used as a control or reference antibody in experiments involving rat models, and is also commonly used in flow cytometry and immunohistochemistry applications.

Can the eFluor Organic fluorochromes be used for intracellular staining?

Yes, the eFluor Organic fluorochromes can be used for intracellular staining. The eFluor organic fluorochromes maintain bright signal and require minimal changes in compensation when fixed with eBioscience IC Fixation Buffer (Cat. No. 00-8222-49) and Permeabilization Buffer (Cat. No. 00-8333-56) or 1-step Fix/Lyse Solution (Cat. No. 00-5333-54, 00-5333-57) (as compared to live cells).

What buffers are compatible with the eFluor Organic fluorochromes and eVolve QDots?

The eFluor Organic fluorochromes and eVolve QDots can be used with flow staining buffers containing PBS and protein.

What is the difference between eFluor Organic Dyes and eVolve Qdots?

The eFluor Organic Dyes (eFluor 450, APC-eFluor 780, PerCP-eFluor 710, eFluor 710) are conventional fluorochromes. In contrast, the eVolve line of products are Quantum dots.

Are the eFluor Organic Dyes photo-labile?

As with other fluorochromes, we recommend minimal exposure to light to maintain optimal signal.
