Selectively differentiate and isolate Yersinia enterocolitica from food products using Thermo Scientific™ Remel™ CIN Agar Base (Dehydrated) and Yersinia CN selective supplement (R45050). This medium was first described in 1979 by Schiemann and contained cefsulodin, novobiocin, Irgasan, bile salts, and crystal violet as selective agents1. Later he substituted bile salts with sodium desoxycholate to further improve growth and recovery of Y. enterocolitica2. CIN Agar is recommended in Bacteriological Analytical Manual and in Compendium of Methods for the Microbiological Examination of Foods for isolation of Yersinia from food products3,4.
Yersinia enterocolitica is recognised as one of the common causes of diarrhoeal disease in humans causing gastroenteritis and mesenteric lymphadenitis5. Infection by the organism results in diarrhoea, malaise, nausea and fever, plus constant abdominal pain over a period of 1-2 days. The organism has also been shown as a cause of polyarthritis, mesenteric adenitis and septicaemia. It is likely that human infections are directly or indirectly derived from animal sources and may be contracted through the ingestion of contaminated food.

- Recommended in USFDA’s Bacteriological Analytical Manual and in Compendium of Methods for the Microbiological Examination of Foods (APHA) for isolation of Yersinia from food products.
- Easy to Differentiate: Mannitol is the fermentable carbohydrate which, in combination with neutral red dye, enables differentiation of enteric gram-negative bacilli.
- Selective: Cefsulodin, novobiocin, crystal violet, sodium desoxycholate, and Irgasan are selective agents which inhibit Escherichia coli, Klebsiella pneumoniae, Proteus mirabilis, and Pseudomonas aeruginosa, as well as gram-positive organisms.
CIN Agar Base contains peptones that provide nitrogen, amino acids, and peptides necessary for bacterial growth. Yeast extract provides B-complex vitamins. Mannitol is the carbohydrate which, in combination with neutral red dye, enables differentiation of enteric gram-negative bacilli. Organisms which ferment mannitol produce a localized pH drop around the colony which is followed by absorption of the neutral red dye. This results in a characteristic “bulls-eye” colony which has a colorless, translucent outer zone and a red center. Colonies of Y. enterocolitica typically develop a “bulls-eye” appearance after 24-48 hours incubation at 25°C. Crystal violet, sodium desoxycholate, cefsulodin, novobiocin and Irgasan are selective agents which inhibit Escherichia coli, Klebsiella pneumoniae, Proteus mirabilis, and Pseudomonas aeruginosa, as well as gram-positive organisms.
