Cultivate anaerobic bacteria using Thermo Scientific™ Remel™ PRAS Cooked Meat Glucose. This medium is an enrichment broth that is recommended by Virginia Polytechnique Institute (VPI) Anaerobe Labortory Manual¹. It supports the growth of spore-forming and non-spore-forming obligate anaerobes and has the capacity to initiate growth of bacteria from minute inocula². Claros et al. found cooked meat medium superior to thioglycollate broth for stock cultures because it maintains the viability of anaerobic bacteria over extended periods of time³. This medium is also referred to as chopped meat medium.

Cultivate anaerobic bacteria using PRAS Cooked Meat Glucose. R.E. Hungate pioneered the development of Pre-reduced Anaerobically Sterilized (PRAS) media, which are processed in a reduced condition and remain reduced up to and after inoculation⁴.

This medium contains minimal salts, beef heart, peptone, and dextrose which provide amino acids and other nutrients necessary for the growth of anaerobes. Solid meat particles create favorable growth conditions for anaerobes due to the reducing action of sulfhydryl groups from muscle protein⁵. Cystine provides sulfahydryl groups which bind hydrogen and produce anaerobic conditions. Vitamin K and hemin are essential for the growth of certain anerobes.
