Why are my PCR tubes crushed/deformed after my PCR run?

The heated thermal cycler lid is designed to make sure that optimized pressure is applied on the microplate for an efficient PCR reaction. When using PCR tubes, excess pressure on the tubes, from the heated lid can cause deformation of the tubes. To avoid this, we recommend using the Tray/Retainer set, as shown below:

- PCR optimization: each block can be set with a specific temperature, which is ideal for precise control over PCR optimization.
- Run more experiments: the three Peltier blocks can also be used to run up to three different annealing temperatures in the same run.

What is the Auto Delta feature on the SimpliAmp Thermal Cycler?

The AutoDelta feature allows you to incrementally increase or decrease the temperature or hold time with each cycle for a given step within a cycling stage. One common application for this is touchdown PCR.

We offer only a 96-well standard (0.2 mL) block that is not interchangeable.

Why does the SimpliAmp Thermal Cycler starter kit contain two different types of trays?

The two different types of trays are for use with different tubes:

-MicroAmp 96-well Tray/Retainer Set for Veriti Systems (Cat. No. 4381850) - this tray is for use with 0.2 mL single tubes without caps (Cat. Nos. N8010533, N8011533), 0.2 mL 8-tube strip with attached dome caps (Cat. No. A30589), and 0.2 mL tube strips without caps (Cat. No. 4316567)
-MicroAmp 96-well Tray for VeriFlex Blocks (Cat. No. 4379983) - this tray is for use with 0.2 mL single tubes with caps (Cat. Nos. N8010540, N8011540)

What is the method storage capacity for the SimpliAmp Thermal Cycler?

The storage capacity for the SimpliAmp Thermal Cycler is approximately 5.9 GB. The run methods and run records stored use most of the memory. It can store 1.2 million run methods.

How well does the SimpliAmp Thermal Cycler perform with a 4 degrees C hold?

The SimpliAmp PCR System can be used with a 4 degrees C hold. Further, our in-house QC testing requires the passing of a “no leak” test make sure that any accumulated water (due to condensation when the block temperature is set to low temperature for a long period of time) will not leak into the instrument causing electronic failure.

Can I purchase the sample block for the SimpliAmp Thermal Cycler (Cat. No. A24811) as a standalone product? Is it replaceable/interchangeable?

Unfortunately, we do not sell the sample block for the SimpliAmp Thermal Cycler separately. Only the ProFlex series and the Automated series of thermal cyclers have interchangeable blocks that we sell separately.

What is the optimal ramp rate for my thermal cycler?

There is no universal ramp rate for a thermal cycler because both the block and sample ramp rates are different for each thermal cycler. The sample ramp rate changes with the volume of the sample since more time and energy are required to heat higher sample volumes. The ramp rate may also need to be changed for a particular PCR assay or primer pair if you are trying to use the same PCR assay on two different thermal cyclers, for example.

As a reference, the block ramp rate commonly used with DreamTaq Polymerase on the MiniAmp thermal cycler is 3.0 C/sec. In a 50 µL PCR reaction, this translates to a sample ramp rate of ~1 C/sec.
