SuperBoost™ EverRed and EverBlue Colorimetric HRP Kits use poly-HRP-mediated tyramide signal deposition for permanent labeling of a target protein in IHC, enabling sensitive and specific colorimetric tissue staining that is wash resistant, water/organic solvent mountable, and archivable.

SuperBoost™ colorimetric signal anchoring is a highly sensitive, permanent method for the detection of targets in multiplexable colorimetric immunohistochemistry (IHC) experiments. SuperBoost™ technology combines the permanent staining of poly-HRP-mediated tyramide signal deposition with the bright colors of SuperBoost EverRed and EverBlue to discern signal from noise, yielding precision and sensitivity in colorimetric tissue staining applications.

Tyramide signal deposition in the Tyramide SuperBoost™ kits uses the catalytic activity of horseradish peroxidase (HRP) for high-density labeling of a target protein, enabling permanent colorimetric tissue staining with SuperBoost EverRed and/or EverBlue that is wash resistant, water/organic solvent mountable, and archivable. Unlike many other colorimetric IHC staining methods, SuperBoost EverRed and EverBlue staining is covalently attached to the target resulting in a sharp and intense chromogenic signal that is maintained without needing to air-dry tissue or use unconventional dehydration/clearing protocols.

The kits are simple to use and easily adapted to standard IHC experimental protocols using any tissue type, including formalin-fixed, paraffin-embedded (FFPE) and cryopreserved tissue. Tissues labeled using the SuperBoost EverRed and EverBlue kits can be imaged using any type of brightfield microscope, producing high-resolution images.

How does SuperBoost color signal anchoring work?
SuperBoost EverRed and EverBlue HRP Kits use a simple workflow similar to standard IHC staining methods. For SuperBoost EverRed and EverBlue HRP tissue staining, an antigen is detected by a primary antibody, followed by staining with a secondary antibody or streptavidin (for biotinylated primary antibodies) conjugated with poly-HRP. HRP then enzymatically activates a chromogen tyramide, resulting in localized covalent deposition of the activated tyramide derivative at tyrosine residues on and surrounding the protein epitope targeted by the primary antibody. Treatment of the labeled tissue with the EverRed or EverBlue reagent then results in a permanent colorimetric signal at the site of the covalent tyramide deposition.

The SuperBoost EverRed/EverBlue HRP Kits, goat anti-mouse IgG/goat anti-rabbit can be used for permanent red or blue colorimetric detection of mouse or rabbit IgG primary antibodies in IHC applications. The kits include the goat anti-mouse IgG or goat anti-rabbit IgG poly horseradish peroxidase (HRP) conjugate, tyramide HRP substrate, and SuperBoost EverRed or EverBlue reagent along with the buffers and reagents needed for tissue staining and color development. Sufficient reagents are provided to stain 150 tissue sections.

Sufficient reagents are provided to stain 150 tissue sections.

SuperBoost EverRed/EverBlue HRP Substrate Kits
The SuperBoost EverRed/EverBlue HRP Substrate Kits include the tyramide HRP substrate and SuperBoost EverRed or EverBlue reagent for permanent red or blue colorimetric detection in IHC applications using your own HRP conjugate. Sufficient reagents are provided to stain 150 tissue sections or 5 x 150 tissue sections.
