Suramin hexasodium salt (CAS 129-46-4) is a polysulfonated naphthylurea derivative that competitively inhibits DNA topoisomerase II an enzyme essential for managing DNA supercoiling and topology during replication and transcription with an IC50 of approximately 5 M Suramin selectively inhibits cellular DNA synthesis and proliferation notably blocking SV40 DNA replication entirely at a concentration of 40 M in HeLa cells The compound demonstrates differential inhibitory effects towards DNA polymerases and (IC50 of 8 M and 36 M respectively) Additionally suramin significantly inhibits bacterial RecA-mediated ATPase activity strand exchange processes and proteolytic cleavage of the LexA transcriptional repressor In preclinical models of pulmonary hypertension suramin reduces vascular remodeling inflammation and pulmonary artery pressure reflecting its utility in cellular proliferation enzyme function and disease model research
