Applied Biosystems™ TaqMan™ Express Endogenous Control Plates use TaqMan™ probe-based chemistry and are designed for use on the suite of Applied Biosystems™ Real-Time PCR Systems – together the gold standard in quantitative gene expression offering the greatest sensitivity, specificity, reproducibility, and the broadest dynamic range.

The first aspect of any experiment looking at relative quantitation of gene expression should be the selection of endogenous control gene(s), to normalize for variations in sample input. The TaqMan™ Express Human Endogenous Control Plate contains 32 genes, plated in triplicate, that have been shown to be good candidates for such an experiment. This collection of genes has been selected from literature searches and/or whole genome microarray tests carried out on numerous human tissues. They have been shown to be expressed constitutively and at moderate abundance across most test samples.

These 96-well plates are pre-configured with the most appropriate TaqMan™ Gene Expression Assays for a specific biological process, pathway, or disease state. Each plate contains predefined assays and endogenous controls dried-down in the wells, ready for accurate assessment of an entire gene signature in one simple experiment. TaqMan Assays (TaqMan probe and PCR primer sets) are considered the gold standard in gene expression analysis, enabling you to achieve high specificity and sensitivity and a broad dynamic range.

How do I obtain TaqMan documentation files for my TaqMan array plates?

Beginning in April 2016, TaqMan documentation files for TaqMan Array Plates (assay information files (AIF), experimental setup files, and plate layout files) can be downloaded from the Thermo Fisher Scientific website at https://www.thermofisher.com/taqmanfiles. To obtain your TaqMan documentation files:
• Select the appropriate product type.
• Enter the information being requested for the product type.
• Fixed TaqMan array plates: Enter the Part number and Batch number
• TaqMan Flexible Content panel array plates/Custom TaqMan array plates: Enter the Sales Order number and Batch number
• Click Submit.

Can poly A+ purified RNA samples be used with the TaqMan Human Endogenous Control Plate (Cta. No. 4309199)?

No. The plate has been designed to evaluate only total RNA. Ribosomal RNA will be removed from poly A+ purified RNA samples, and is required for accurate detection of the 18s rRNA endogenous control assay used on the plate.

What types of templates can be used with the TaqMan Human Endogenous control Plate (P/N 4396840)?

The plate amplifies mRNA contained within total RNA. Non-human templates are not recommended, as all the assays are human specific with the exception of 18S rRNA and the internal positive control. Additionally, Poly A+ RNA cannot be accurately evaluated by the 18S rRNA assay as most of the rRNA has been removed.
