TEV protease is a highly specific cysteine protease with exceptional catalytic efficiency It recognizes the sequence ENLYFQ S but the amino acid at the P1 position can also be G A M C or H (1) TEV protease is commonly used to remove affinity purification tags from fusion proteins such as Glutathione S-transferase (GST) His or other tags As TEV protease itself carries a His-tag it can easily be removed by nickel affinity resin after the proteolysis reaction It is suitable for cleavage of His-tagged fusion proteins since incompletely cleaved His-tagged recombinant protein and the removed His-tagged portion along with His-tagged TEV protease itself can all be bound and removed through nickel column chromatography The desired target protein is obtained in the column flow-through in the meanwhile Overnight digestion at 4 with TEV protease is recommended TEV protease retains activity within pH 6 0-9 0 and is significantly reduced or even lost at pH 5 0 or below
