Mouse Monoclonal Antibody

Description: The eBio1A6 monoclonal antibody reacts with human thymic stromal-derived lymphopoietin receptor (TSLPR). TSLPR is an approximately 50 kDa protein with significant similarity to the common gamma-chain. TSLPR complexes with IL-7R alpha (CD127) to form the high affinity receptor that binds thymic stromal-derived lymphopoietin (TSLP). Human TSLPR is expressed by monocytes and CD11c+ dendritic cells, and TSLP binding induces the expression of the Th2 cell-attracting chemokines CCL17 and CCL22. Furthermore, the TSLPR-induced activation of dendritic cells indirectly results in the increased secretion of Th cytokines IL-4, -5 and -13, which may be necessary for the regulation of CD4+ T cell homeostasis. In mice, deficiency of TSLPR has no effect on lymphocyte numbers, whereas double deficiency of TSLPR and common gamma-chain results in fewer lymphocytes than seen in mice deficient in the common gamma-chain alone. The eBio1A6 monoclonal antibody is able to cross-block binding of another anti-human TSLPR monoclonal antibody, 1D8. Applications Reported: This eBio1A6 (1A6) antibody has been reported for use in flow cytometric analysis. Applications Tested: This eBio1A6 (1A6) antibody has been pre-titrated and tested by flow cytometric analysis of TSLPR transfected cells. This can be used at 5 μL (0.125 μg) per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empir...

Thymic stromal lymphopoietin (TSLP) has recently been identified as an important factor capable of driving dendritic cell maturation and activation. It is involved in the positive selection of regulatory T cells, maintenance of peripheral CD4+ T cell homeostasis and the induction of CD4+ T cell-mediated allergic reaction. TSLP is also capable of supporting the growth of fetal liver and adult B cell progenitors and their differentiation to the IgM-positive stage of B cell development. Its receptor TSLP-R will bind TSLP in a low-affinity fashion in transfected cells; co-transfection with IL-7R-alpha cDNA results in high-affinity binding and a functional heteromeric complex. This heteromeric receptor requires stat5 for TSLP-mediated signal transduction and is inhibited by SOCS-1.

I am unable to analyze my cells stained with eFluor Organic Dyes today. What options do I have?

Our options will depend on the samples you are analyzing.

If cell viability is not critical, you can store your stained samples at 4 degrees C or on ice overnight in the dark and analyze the following day.

For samples stained with eFluor organic fluorochromes, we recommend that cells be suspended in 100 uL of Flow Cytometry Staining Buffer (Cat. No. 00-4222) and 100 uL of eBioscience IC Fixation Buffer (Cat. No. 00-8222); samples can be incubated for up to 3 days at 4 degrees C in the dark. Alternatively, the 1-step Fix/Lyse Solution (Cat. No. 00-5333) can be used. This is a great option when working with whole blood but also works for other cell types.

Can the eFluor Organic fluorochromes be used for intracellular staining?

Yes, the eFluor Organic fluorochromes can be used for intracellular staining. The eFluor organic fluorochromes maintain bright signal and require minimal changes in compensation when fixed with eBioscience IC Fixation Buffer (Cat. No. 00-8222-49) and Permeabilization Buffer (Cat. No. 00-8333-56) or 1-step Fix/Lyse Solution (Cat. No. 00-5333-54, 00-5333-57) (as compared to live cells).

What buffers are compatible with the eFluor Organic fluorochromes and eVolve QDots?

The eFluor Organic fluorochromes and eVolve QDots can be used with flow staining buffers containing PBS and protein.

What is the difference between eFluor Organic Dyes and eVolve Qdots?

The eFluor Organic Dyes (eFluor 450, APC-eFluor 780, PerCP-eFluor 710, eFluor 710) are conventional fluorochromes. In contrast, the eVolve line of products are Quantum dots.
