Life Science Buffers
Balanced salt solutions and powders used for molecular, protein, nucleic acid, and cell biology applications including cell culture, electrophoresis, ELISA, and chromatography processes; available in a variety of formulations and grades.
Buffer solutions serve to keep pH (acidity or alkalinity) nearly constant in a variety of chemical and biological applications. For example, a bicarbonate buffer helps maintain the pH of blood, a buffered saline maintains cellular contents at a consistent pH level, and buffers help maintain a narrow pH range for enzymes to function correctly.
Most biological samples used in research are kept at a pH of about 7.4. Common biological buffers used for tissue culture include Dulbeccos phosphate buffered saline (PBS), Tris base, HEPES, MOPS, PIPES, and other formulations developed for specific cell lines or applications. Most buffers used in cell culture are also DNase-, RNase-, and protease-free.
The choice of buffer for a particular biological reaction or site depends on several factors:
- Temperature
- Desired or target pH
- Buffer toxicity (to the system)
- Buffer interactions with other system components

Calcein AM/PI Live-Dead Cell Staining Kit is a reagent kit used to distinguish between viable mammalian cells with esterase activity and dead cells The kit contains two probes Calcein AM and Propidium Iodide (PI) Calcein AM is a probe that stains live cells it is a derivative of Calcein with an added AM moiety which makes it more hydrophobic and allows better cell permeability Calcein AM itself is non-fluorescent but once it enters live cells it is hydrolyzed by intracellular esterases into Calcein which can emit strong green fluorescence PI is a probe for dead cells which enters only cells with compromised membranes staining them red The fluorescence intensities of these two probes reflect cell viability and toxicity The kit is easy to use 30 minutes of staining incubation After staining results can be detected using fluorescence microscopy fluorescence microplate readers or flow cytometry This kit is unsuitable for bacteria or fungi

The AO/PI Double Staining Kit is a reagent kit enabling rapid detection of cell viability This kit provides two nuclear dyes Acridine Orange (AO) and Propidium Iodide (PI) AO can penetrate intact cell membranes and stain viable cells green During apoptosis chromosomes become condensed or fragmented into differently sized segments In this scenario AO stains apoptotic cells densely with yellow-green fluorescence or as yellow-green fragments The other nuclear dye PI cannot pass through intact cell membranes therefore it does not stain viable or apoptotic cells in red but stains cells with compromised membranes (e g necrotic cells) When using these two dyes in combination viable cells exhibit green fluorescence apoptotic cells show dense varied-size yellow-green or orange fluorescence and necrotic cells exhibit strong red fluorescence

Annexin V is a Ca2 dependent phospholipid-binding protein that specifically binds to phosphatidylserine (PS) Normally PS resides on the inner surface of the cell membrane s phospholipid bilayer During early stages of apoptosis it translocates from the inner leaflet to the outer surface thus exposing itself to the extracellular environment Cy5-labeled Annexin V can serve as a fluorescent probe to detect apoptosis using flow cytometry or fluorescence microscopy within 10-20 minutes SYTOX Green is a non-cell-permeable cyanine fluorescent dye and stains only dead cells It can be used effectively to differentiate normal viable cells from necrotic cells For cells at late apoptotic stages or dead cells SYTOX Green can penetrate the disordered regions of cell membrane reach the cell nucleus embedding into the DNA double helix to form a SYTOX Green-DNA complex resulting in a 500-fold increase of fluorescence intensity with bright green fluorescence

When extracting secreted proteins from culture medium in vitro protein production ceases and degradation increases Cell crude extracts contain many endogenous enzymes such as phosphatases and proteases capable of degrading proteins in the extracts The best way to improve the yield of intact proteins is to add inhibitors targeting these known enzymes The Protease Inhibitor Cocktail is used in tissue culture media to enhance protein stability It functions by inhibiting proteases responsible for the degradation of both phosphorylated and non-phosphorylated protein substrates This cocktail is provided as a supplement for tissue culture media to prevent degradation of secreted proteins This Protease Inhibitor Cocktail has been optimized and tested specifically for tissue culture media This protease inhibitor cocktail is provided in DMSO as a ready-to-use solution

Under stable environmental conditions endogenous protein synthesis and degradation are balanced thus maintaining stable cellular protein levels Cell crude extracts contain numerous endogenous enzymes such as phosphatases and proteases that can degrade proteins in the extracts During in vitro protein extraction from cells and tissues protein synthesis practically ceases while protein degradation increases The optimal way to enhance yields of intact proteins is to add inhibitors for these known enzymes The Protease Inhibitor Cocktail for bacterial cell extracts (100X in DMSO) is designed for bacterial extracts to enhance the stability of proteins This cocktail inhibits proteases responsible for degrading non-phosphorylated or phosphorylated protein substrates This Protease Inhibitor Cocktail is provided as a ready-to-use solution and it is formulated in DMSO

The MOPS Buffer [3- (N-morpholino)propanesulfonic acid buffer] is a versatile zwitterionic Good's buffer used in biological research. With a pKa of 7.20, MOPS is an excellent buffer for many biological systems at near-neutral pH, and maintains a pH range from 6.5 to 7.9. It is often used as a buffering agent in biology and biochemistry, and is recommended for polyacrylamide gel electrophoresis, as a running buffer, in separating RNA in agarose gels, and for protein purification chromatography.
