PAN-Biotech 1x Erythrocyte Lysis Buffer (1x RBC Lysis Buffer), 6 x 100 ml. DESCRIPTION: In normal whole-blood, erythrocyte concentration is 1000x higher than leukocyte concentration, making erythrocytes an impediment to investigation or analysis of leucocytes. Erythrolyse, or red blood cell lysing buffer is intended for the lysis of red blood cells in samples with white blood cells as for example EDTA-/citrate/heparin-treated whole-blood, Buffy Coats (PBMCs, leucocyte films), and bone marrow. This buffer contains ammonium chloride, which lyses red cells, with minimal effect on lymphocytes when used as instructed. Nucleated red cells are not effectively lysed with ammonium chloride. COMPOSITION: Contains ammonium chloride, potassium hydrogencarbonate, and EDTA. Erythrocyte Lysis Buffer (1x) is sterile filtered (0.2 μm) and contains no fixative and no preservatives. CFSE is a reagent useful for cell tracking, proliferation studies, and cell motility studies. After passively diffusing into cells, it is converted to a fluorescent carboxyfluorescin molecule as its acetate groups are cleaved by intracellular esterases. The fluorescent CFSE is retained inside the cell for extended periods, because the molecules forms stable covalent crosslinks with the intracellular proteins. At each cell division, the intensity of the CFSE fluorescence is halved. The peak excitation of the molecule is 494 nm and the peak emission is 521 nm and it may be used with standard fixatives containing formaldehyde and permeabilization buffers that contain saponin.
