ODN 2006 (Type B) Endotoxin-free (sterile), AdipoGen Life Sciences. Chemical. MW: 7,720g/mol. Unmethylated CG dinucleotides within particular sequence contexts are responsible for the immunostimulatory activity of bacterial DNA. Synthetic oligonucleotides (ODN) that contain such CpG motifs (CpG ODNs) mimic microbial DNA. The innate immune system of vertebrates has the ability to recognize CpG motifs in microbial DNA via the Toll-like receptor (TLR) 9 if the CpG ODN were free of additional immune stimulatory contaminants often present in synthetic commercial CpG ODN preparations designed for molecular biology applications (i.e. PCR).

In recent years several groups have studied the sequence requirements, specificity, signaling pathways and kinetics of the TLR (Toll-like receptor) 9 suppression by inhibitory oligonucleotide motifs, which led to a class of novel inhibitory oligonucleotide (iODNs), that is independent of the previously thought species preference. Subsequently it has been discovered that telomeric DNA repeats (TTAGGG)n can block immune activation by CpG-ODNs. Short, 11-15 base long oligonucleotides were synthesized that were capable of potently inhibiting CpG-stimulation.

DNA Ligase I maintains the major DNA Ligase activity in proliferating cells by joining Okazaki fragments during lagging strand DNA replication. Human DNA Ligase I also has an essential role in DNA repair pathways, where it catalyzes the formation of phosphodiester bonds between adjacent 5' phosphoryl and 3' hydroxy termini at single breaks in duplex DNA molecules. In addition, DNA Ligase I plays a role in sealing nicks during excision repair. Similar to other DNA ligases, DNA Ligase I is built around a common catalytic core. Increased levels of DNA Ligase I are found in human tumors, as compared to benign tissues, as well as in peripheral blood lymphocytes. DNA Ligase I activity is altered in the chromosomal breakage deficit Bloom's syndrome (BS). Individuals with BS either have decreased levels of abnormally thermolabile DNA Ligase I or possess a dimeric form of this enzyme. Recognizes a cell surface glycoprotein of 95/115/135 kDa (depending upon the extent of glycosylation), identified as CD43. 70-90% of T-cell lymphomas and from 22-37% of B-cell lymphomas express CD43. No reactivity has been observed with reactive B-cells. So a B-lineage population that co-expresses CD43 is highly likely to be a malignant lymphoma, especially a low-grade lymphoma, rather than a reactive B-cell population. When CD43 antibody is used in combination with anti-CD20, effective immunophenotyping of the lymphomas in formalin-fixed tissues can be obtained. Co-staining of a lymphoid infiltrate with anti-CD20 and anti-CD43 argues against a reactive process and favors a diagnosis of lymphoma. Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. CF Dyes offer exceptional brightness and photostability.
