Thiosulfate Citrate Bile Salts Sucrose (TCBS) Agar is used for the selective isolation of Vibrio cholerae and other enteropathogenic vibrios according to ISO 21872-1:2017 and ISO 21872-2:2007. Thiosulfate Citrate Bile Salts Sucrose (TCBS) Agar is not intended for use in the diagnosis of disease or other conditions in humans. TCBS Agar is also called Vibrio Selective Agar, and prepared according to the formula of Kobayashi et al. This formula is a modification of the selective medium from Nakanishi. All pathogenic Vibrio spp., except Vibrio hollisae, will grow on TCBS Agar. EC Medium w/ MUG is used for the fluorogenic detection of Escherichia coli in a laboratory setting. Muller-Kauffmann Tetrathionate-Novobiocin Broth is used for the selective and differential isolation of Salmonella spp. in a laboratory setting and is included in ISO 6579:2017. This medium is not intended for use in the diagnosis of disease or other conditions in humans. A selective enrichment medium for the detection of Salmonella spp. in food, animal feed and in environmental samples from the food production area as described in ISO 6579-1:2017. Mueller originally described a tetrathionate broth that inhibited lactose fermenting Enterobacteriaceae but not Salmonella. Kauffmann later modified the formulation to include ox bile and brilliant green to improve selectivity. Finally, Jeffries added Novobiocin to inhibit Proteus species. NEOGEN YM Broth, 5 kg
YM Broth is used for the cultivation of fungi in a laboratory setting. YM Broth is not intended for use in the diagnosis of disease or other conditions in humans. YM Broth is prepared according to the formulation published by Wickerham. Wickerham suggested that YM Broth, acidified to pH 3.0 – 4.0, be used as an enrichment medium for yeasts from populations also containing bacteria and molds. To favor isolation of fermentative species, add a layer of sterile paraffin oil 1 cm deep on the surface of the inoculated broth. Cultures should be incubated until growth appears, and then streaked onto YM Agar to obtain isolated yeast colonies. To isolate fermentative and oxidative strains, place acidified inoculated YM Broth on a rotary shaker for 1 to 2 days. This favors yeast recovery while preventing sporulation of molds. Eosin Methylene Blue Agar Levine is used for the isolation and differentiation of Gram-negative enteric bacilli and is not intended for use in the diagnosis of disease or other conditions in humans.This medium was introduced in 1916 by Holt-Harris and Teague to differentiate Escherichia spp. and Aerobacter spp. It was modified by Levine in 1918 who removed sucrose from the formula and increased the lactose content. The distinctive metallic sheen produced by E. coli on this medium is due to acid production resulting in an amide bonding between the eosin and methylene blue, other coliforms do not produce enough acid to cause this reaction. DC Medium with BCIG is used in the chromogenic differentiation of E. coli from other coliforms in water samples using the membrane filtration method in a laboratory setting. This medium is enhanced by the addition of a chromogenic agent, BCIG, 5-bromo-4-chloro-3-indolyl-ß-D-glucuronide to detect glucuronidase activity. The presence of the enzyme ß-D-glucuronidase differentiates most E. coli spp. from other coliforms, and is the same enzyme used in the MUG reaction. Rochaix first noted the value of esculin hydrolysis in the identification of enterococcci. Meyer and Schonfeld added bile to the esculin medium and demonstrated 61 of 62 enterococci strains were able to grow and hydrolyze esculin, while other streptococci could not.Molecular taxonomic studies of the genus Streptococcus have placed enterococci, previously described as group D streptococci, in the genus Enterococcus.
