The Ready-to-Assay MRGPRD Receptor Frozen Cells are a simple, thaw-and-assay cell format for use in studying drug candidates (small molecules or biologics) targeting GPCRs. This assay assesses ligand (e.g. β-alanine) based activation of MRGPRD GPCR activity via detection of increasing levels of intracellular calcium. The included cell line overexpresses MRGPRD alongside endogenous high levels of a promiscuous G-protein, coupling the receptor to the calcium signaling pathway. Upon activation of MRGPRD, the resulting calcium activity can be fluorescently assayed. Ready-to-assay kits consist of two vials of the stably modified clonal cell line in a convenient thaw-and-use format. The ChemiScreen CGRP1 Calcitonin Receptor-Like Receptor Stable Cell Line is an engineered cell line for use in studying drug candidates (small molecules or biologics) targeting GPCRs. This cell line enables assessment of ligand (e.g. calcitonin) based activation of CALCRL - RAMP1 GPCR activity via detection of increasing levels of intracellular calcium. The included cell line overexpresses CALCRL alongside RAMP1 modifying protein and endogenous high levels of the promiscuous G protein, Gα15, coupling the receptor to the calcium signaling pathway. Upon activation of CALCRL, the resulting calcium activity can be fluorescently assayed. This stable modified cell line is provided as two vials of cryopreserved cells. ChemiScreen membrane preps can be used to screen for ligand binding affinities using a competitive radio-labeled ligand approach. Increased binding affinities will lead to higher radioactive signal. Membrane preps contain 1 mL of 1 mg/mL frozen membranes derived from a stably modified clonal cell line which was chosen for optimal receptor expression. ATCC Canine distemper virus; Strain: Snyder Hill
Canine distemper virus strain Snyder Hill was derived from ATCC VR-526 by adaptation to growth in Vero cells (ATCC CCL-81). ATCC VR-526, deposited by L.E. Carmichael, was isolated from the brain of a dog with clinical distemper in September 1950. Because it was propagated in animal tissue, ATCC will no longer offer that product.

This cell line enables assessment of compound-engagement-based stabilization of PYK2 via detection of PYK2 levels. This product uses EFC technology. The cell line included overexpresses PYK2 target protein fused to ProLabel. In the absence of compound that binds and stabilizes the target, this protein is denatured by the heat treatment and no EFC signal is produced. When compound engages and heat-stabilizes the target, EA contained in the detection reagent produces EFC signal. This stable modified cell line is provided as two vials of cryopreserved cells.
