The Ready-to-Assay BB3 Bombesin Receptor Frozen Cells are a simple, thaw-and-assay cell format for use in studying drug candidates (small molecules or biologics) targeting GPCRs. This assay assesses ligand (e.g. bombesin) based activation of BRS3 GPCR activity via detection of increasing levels of intracellular calcium. The included cell line overexpresses BRS3 alongside endogenous high levels of a promiscuous G-protein, coupling the receptor to the calcium signaling pathway. Upon activation of BRS3, the resulting calcium activity can be fluorescently assayed. Ready-to-assay kits consist of two vials of the stably modified clonal cell line in a convenient thaw-and-use format. VTCN1 contains two Ig-like V-type (immunoglobulin-like) domains. The expression of VTCN1 is up-regulated by IL6 and IL10 and is inhibited by GM-CSF and IL4 on antigen-presenting cells (APCs). VTCN1 / B7-H4 negatively regulates T-cell-mediated immune response by inhibiting T-cell activation proliferation cytokine production and development of cytotoxicity.
