One of a series of pBluescript-based centromere vectors (ATCC#77142-77145, 77157-77158) differing in the yeast selectable marker gene. A YC-type centromere vector permitting visual detection of recombinants and production of ssDNA in E. coli. It contains promoters for in vitro RNA synthesis, priming sites useful for sequencing, and encodes the lacZ alpha (lacZ?) peptide. pRSS56, constructed by ligating a PvuI fragment (bp 498-2412) of pBluescript KS+ to a PvuI fragment (bp 2850-730) of pBS(+), contains the KS MCS from pBluescript KS+ and the unique NdeI and AatII sites between bla and f1 origin of pBS(+). A fragment (1.184 kb) containing the HIS3 gene was inserted into the NdeI site and a cassette contiaing CEN6 and the ARS associated with histone 4 (ARSH4) was inserted into the AatII site of pRSS56. All ends were blunted.

The PathHunter C3AR1 cell line is a stable, engineered cell line for use in studying drug candidates (small molecules or biologics) targeting GPCRs. This cell line enables assessment of ligand (e.g. C3a) based activation of C3AR1 GPCR activity via detection of β-arrestin recruitment. The included cell line overexpresses ProLink-tagged C3AR1 and EA-tagged β-Arrestin-2. Activation of C3AR1 stimulates the recruitment of β-Arrestin-2 and produces EFC signal. The KILR Daudi cell pool is a stable, engineered target cell pool, which closely reflects the heterogeneous native protein expression of tumor environments, used to measure direct cell death mediated by immunooncology treatments (e.g. checkpoint modulators, bispecifics, CAR-T, etc.) via Antibody Dependent Cell-mediated Cytotoxicity (ADCC), Complement Dependent Cytotoxicity (CDC), and Antibody Dependent Cell Phagocytosis (ADCP). This product uses EFC technology. The cell pool is a heterogeneous population of Daudi cells that overexpress a ProLabel-tagged housekeeping gene. Upon effector cells-mediated cell lysis, ProLabel is released to the media, where EA in the detection kit produces an EFC signal proportional to the level of ProLabel released. This allows for calculation of percent lysis of the KILR cell pool. This stable cell pool is provided as two vials of cryopreserved cells.
