Biotium Cytokeratin 8 (KRT8)(KRT8/2115), CF647 conjugate, 0.1mg/mL
Cytokeratin 8 (CK8) belongs to the type II (or B or basic) subfamily of high molecular weight cytokeratins and exists in combination with cytokeratin 18 (CK18). CK8 is primarily found in the non-squamous epithelia and is present in majority of adenocarcinomas and ductal carcinomas. It is absent in squamous cell carcinomas. Hepatocellular carcinomas are defined by the use of antibodies that recognize only cytokeratin 8 and 18. CK8 exists on several types of normal and neoplastic epithelia, including many ductal and glandular epithelia such as colon, stomach, small intestine, trachea, and esophagus as well as in transitional epithelium. Anti-CK8 does not react with skeletal muscle or nerve cells. Epithelioid sarcoma, chordoma, and adamantinoma show strong positivity corresponding to that of simple epithelia (with antibodies against CK8, CK18 and CK19). Reportedly, anti-CK8 is useful for the differentiation of lobular ('ring-like, perinuclear') from ductal ('peripheral-predominant')

Recognizes a protein of 110 kDa, identified as Excision Repair Cross Complementing 1 (ERCC1). It is a mammalian nucleotide excision repair (NER) enzyme involved in repair of damaged DNA. ERCC1 is a homologous to RAD10 in Saccharomyces cerevisiae, which is required in mitotic intrachromosomal recombination and repair. ERCC1 is required in repair of cisplatin-induced DNA adducts and ultraviolet (UV)-induced DNA damage. High expression of ERCC1 has been linked to tumor progression in a variety of cancers including non-small cell lung cancer (NSCLC), squamous cell carcinoma of the head, ovarian cancer and esophageal cancer. The protein encoded by this gene is a receptor for interleukin 7 (IL7). The function of this receptor requires the interleukin 2 receptor, gamma chain (IL2RG), which is a common gamma chain shared by the receptors of various cytokines, including interleukins 2, 4, 7, 9, and 15. This protein has been shown to play a critical role in V(D)J recombination during lymphocyte development. Defects in this gene may be associated with severe combined immunodeficiency (SCID). BarX1 is a member of the Bar subclass of homeobox transcription factors. Studies of the Mouse and chick homolog suggest the encoded protein may play a role in developing teeth and craniofacial mesenchyme of neural crest origin. The protein may also be associated with differentiation of stomach epithelia. Down-regulation of Barx1 promotes hepatocellular carcinoma cell (HCC) migration, invasion and metastasis, whereas up-regulation of Barx1 inhibits HCC migration, invasion and metastasis. This antibody recognizes a protein of 75 kDa, identified as Thrombomodulin. Thrombomodulin is a transmembrane glycoprotein with natural anticoagulant properties. It is normally expressed by a restricted number of cells, such as endothelial and mesothelial cells. In addition, synovial lining and syncytio-trophoblasts of placenta also express thrombomodulin. This protein is present in almost all of benign vascular tumors and majority of malignant vascular tumors (Kaposi's sarcoma, angiosarcoma, and epithelioid hemangioendothelioma). Hence, anti-thrombomodulin serves as a sensitive marker for lymphatic endothelial cells and their tumors. This MAb recognizes a protein of 56.5 kDa, identified as cytokeratin 10 (CK10). CK10 is expressed in all suprabasal layers of the epidermis. In the epidermis, expression of CK10 strictly parallels the extent of differentiation; it is absent in the basal layer, appears in the first suprabasal layers and increases in concentration towards the granular layer. However, CK10 is rarely detected in early stages of vulvar squamous carcinomas (tumors less than 2 cm, clinical stage I) regardless of the tumor grade. In larger and more advanced tumors (greater than 2 cm, clinical stages II and III), CK10 is detected very frequently. CD6 is a type I transmembrane glycoprotein that contains a 24-amino acid signal sequence, three extracellular scavenger receptor cysteine-rich (SRCR) domains, a membrane-spanning domain and a 44-amino acid cytoplasmic domain. The CD6 glycoprotein is tyrosine phosphorylated during TCR-mediated T cell activation. CD6 shows significant homology to CD5. CD6 is present on mature thymocytes, peripheral T cells and a subset of B cells. Recognizes an integral membrane glycoprotein of 150 kDa, identified as CD13 (also known as aminopeptidase-N). The antibody recognizes an extracellular epitope. The CD13 antigen is present on most cells of myeloid origin including granulocytes, monocytes, mast cells, and GM-progenitor cells. It is also expressed by the majority of AML, CML in myeloid blast crisis, and in a smaller fraction of lymphoid leukemias. CD13 is absent from normal lymphocytes, platelets and erythrocytes. The intestine-specific transcription factors CDX1 and CDX2 are important for directing intestinal development, differentiation, proliferation and maintenance of the intestinal phenotype. CDX2 protein expression has been seen in GI carcinomas.
