This kit includes two apoptosis markers, NucView 488 Caspase-3 Substrate (green) and CF594 conjugated Annexin (red), for profiling apoptotic cells with respect to two important apoptosis events, caspase-3 activation and phosphatidylserine (PS) translocation, by flow cytometry or fluorescence microscopy.Unlike traditional fluorogenic caspase substrates that inhibit caspase activity or require cell lysis, NucView 488 a novel fluorogenic caspase-3 substrate, is non-inhibitory to caspases, and permits apoptosis detection within individual intact cells, in real-time. The non-fluorescent substrate is cleaved by caspase-3/7 in apoptotic cells releasing a high-affinity DNA dye that migrates to the nucleus and emits bright green fluorescence, allowing caspase-3/7 detection and visualization of apoptotic nuclear morphology. Exposed PS on apoptotic cells is detected by Annexin V conjugated to CF594 (Abs/Em: 593/614 nm), a bright and photostable deep red dye spectrally similar to Texas Red.
