Terminal labeling is an ideal procedure for the labeling of oligonucleotides with haptens such as biotin or fluorescein. An oligonucleotide can be synthesized using standard, commercially available reagents and labeled after synthesis in a simple and reproducible enzyme reaction.The BIO-PROBE 3'-Oligonucleotide Labeling Kit with Fluorescein-12-ddUTP contains all reagents required for terminal labeling of oligonucletides with fluorescein except the oligonucleotides to be labeled. The BIO-PROBE 3'-Oligonucleotide Labeling Kit provides maximum flexibility for labeling oligonucleotides approximately 15 to 100 nucleotides in length. A standard reaction will label up to 100 picomoles (1 µg of a 30-mer oligonucleotide sequence). A complete kit contains two component packs that are available separately: a Reagent Pack and a Fluorescein-12-ddUTP Dideoxynucleotide Pack. Both packs contain sufficient reagents for 25 labeling reactions. Applications: Southern Blot, Northern Blot, ISH , Dot Blot.

The CYTO-ID Red Long-Term Cell Tracer Kit uses proprietary noncovalent cell labeling technology to stably incorporate a red fluorescent dye containing hydrophobic aliphatic chains into the cell membrane’s lipid bilayer. The dye may be loaded into cells by following the included protocol. The labeling buffer is isotonic for mammalian cells and contains no detergents or organic solvents. The appearance of labeled cells may vary depending upon the cell type from uniformly bright to punctuate. This difference is thought to relate to the extent of membrane internalization occurring after cell labeling. The CYTO-ID Red Tracer Dye fluorescence is independent of pH within normally encountered physiologic ranges and fluorescence intensity per cell is typically unaffected by the ultimate pattern of dye distribution. HiLyte Fluor* 555 Labeling Kit-NH2 is used mainly for the preparation of red fluorescence-labeled proteins, such as IgG, for immunostaining, and cellular proteins for tracing. NH2-reactive HiLyteFluor 555, a component of this kit, has a succinimidyl group (NHS) that reacts with amino groups on proteins or other molecules (Fig. 1). This kit contains all the reagents necessary for labeling. Each tube of HiLyte Fluor 555 can label up to 200 μg of IgG, conjugating about 4 to 6 HiLyte Fluor 555 molecules per IgG molecule. The labeling process is simple-add the NH2-reactive HiLyte Fluor 555 to IgG solution on a membrane and incubate at 37ºC for 10 minutes. The excess HiLyte Fluor 555 molecules can be removed by a filtration tube.
