The cells overexpress wild type MCHR1, a naturally Gi/Go coupled GPCR. Dojindo Molecular Technologies Inc Liperfluo(50UGx5) is used to monitor lipid peroxidation in ferroptosis research, emitting intense fluorescence by a lipid peroxide specific oxidation, and is applicable for microscopy and FCM analysis
Liperfluo is designed and exclusively developed by Dojindo for detection of lipid peroxides. Among fluorescent probes that detect Reactive Oxygen Species (ROS), Liperfluo is the only compound that can specifically detect lipid peroxides. Since the excitation and emission wavelengths of the oxidized Liperfluo are 524 nm and 535 nm, respectively, both photo-damage and auto-fluorescence can be minimized. The tetraethyleneglycol group linked to one end of the diisoquinoline ring helps its solubility and dispersibility to aqueous buffer. Though Liperfluo's oxidized form is almost nonfluorescent in an aqueous media, it emits fluorescence in lipophilic sites such as cell membranes. Therefore, it can easily be applied to lipid peroxide imaging by a fluorescence microscopy and a flow cytometric analysis for living cells. Dojindo Molecular Technologies Inc DNA DAMAGE QUANTIFIC KIT (20 SAMPLES) is a colorimetric microplate assay and can determine the number of abasic sites in genomic DNA samples.
Oxidative damage to DNA is a result of its interaction with reactive oxygen species (ROS), in particular the hydroxy radical. Hydroxy radicals, which are produced from superoxide anion and hydrogen peroxide by the Fenton reaction, produce multiple modifications in DNA. Oxidative attacks by hydroxy radicals on the deoxyribose moiety leads to the release of free bases from DNA, generating strand breaks with various sugar modifications and simple abasic sites (AP sites). In fact, AP sites are one of the major types of damage generated by ROS. The Aldehyde Reactive Probe reacts specifically with an aldehyde group present on the open ring form of the AP sites. DNA Damage Quantification Kit contains all the necessary solutions for detecting between 1 to 40 AP sites per 1 x 105 base pairs.
