The PathHunter VIPR1 Activated GPCR internalization eXpress kit provides an easy to-use cell based assay to study drug candidates (small molecules or biologics) affecting GPCR activation and internalization. This kit uses EFC technology to measure ligand-induced internalization of VIPR1 via detection of localization to the early endosome. The included cell line overexpresses EA-tagged β-Arrestin-2 as well as a ProLink-tagged early endosome protein and untagged VIPR1. Upon ligand-induced receptor activation, β-Arrestin-2 is recruited to the activated GPCR and the resulting VIPR1-arrestin complex is internalized to the early endosome, leading to production of EFC signal. eXpress kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, detection reagent, and assay plates. SensoLyte Green Glutaminyl Cyclase Activity Assay *Fluorimetric*, 1 Kit, The SensoLyte Green Glutaminyl Cyclase Activity Assay Kit provides a convenient, two-step homogeneous procedure for measuring enzyme activity from various sources using a green fluorescence substrate. During the first step, substrate is incubated with Glutaminyl Cyclase or enzyme-containing samples and converted into the pyroglutamate form. Secondly, glutaminyl cyclase developer is added to remove pGlu residue and generate the green fluorophore. It can be detected with excitation at 490 nm and emission at 520 nm. Produced fluorescence is proportional to the enzyme activity. This kit is ideal for high throughput screening (HTS) of glutaminyl cyclase activators and inhibitors. The long wavelength of the green fluorophore is less interfered by autofluorescence of cell components and test compounds. Assay sensitivity is 1.5 ng/mL of active enzyme.
