The cAMP Hunter RXFP1 eXpress kit provides an easy-to-use cell based assay to study drug candidates (small molecules or biologics) targeting GPCRs. This kit assesses ligand (e.g. relaxin-2) based activation of RXFP1 GPCR activity via detection of increasing levels of intracellular cAMP. This product uses EFC technology. The cells overexpress wild type RXFP1, a naturally Gs coupled GPCR. Upon activation of RXFP1, the increase in levels of intracellular cAMP leads to an increased signal from EFC when the cAMP detection kit is applied. eXpress kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, detection reagent, and assay plates. Cayman Chemical cPLA2 asy KIt 96Wel
Arachidonoyl thio-PC is a substrate for cPLA2 by virtue of the presence of arachidonic acid at the sn-2 position of the glycerophospholipid. Hydrolysis of the arachidonoyl thioester bond at the sn-2 position by PLA2 releases free thiol which can be detected by DTNB. This assay can be used to determine the activity of cPLA2 in purified preparations, cell cultures, or tissue homogenates that are known to contain only cPLA2. Use of this assay with preparations containing more than one type of PLA2 will result in the measurement of total PLA2 activity rather than cPLA2 alone. Isozyme-specific cPLA2 activity can be measured by excluding sPLA2 or inhibiting iPLA2 activities in the assay.
