Cell Biolabs’ HDL and LDL/VLDL Cholesterol Assay Kit is a simple fluorometric assay that can measure the amounts of HDL and LDL/VLDL cholesterol present in plasma or serum samples within a 96-well microtiter plate format. The kit provides reagents for separating and quantifying HDL and LDL/VLDL cholesterol. The assay will detect total cholesterol (cholesteryl esters plus free cholesterol) in the presence of cholesterol esterase or only free cholesterol in the absence of the esterase enzyme. Each kit provides sufficient reagents to perform up to 192 assays, including blanks, cholesterol standards and unknown samples. Sample cholesterol concentrations are determined by comparison with a known cholesterol standard. Cholesteryl esters can be quantified by subtracting the free cholesterol values from the total cholesterol value.

Cayman’s Superoxide Dismutase Assay Kit provides a convenient method of measuring SOD activity in plasma, serum, erythrocyte lysates, tissue homogenates, and cell lysates. The assay relies on xanthine oxidase to produce superoxide radicals, which then reduce a tetrazolium salt to a formazan dye, detectable at 450 nm. SOD in the sample inhibits this reduction by catalyzing the dismutation of superoxide radicals. Thus, the degree of inhibition is directly proportional to SOD activity. The SOD Assay Kit measures all three types of SOD (Cu/Zn-, Mn-, and Fe-SOD). Mitochondrial Mn-SOD can be assayed separately following the procedure outlined in the kit booklet.

Cayman’s HDAC Cell-Based Assay Kit provides a robust and EAsy-to-use platform for studying HDAC activity modulators in whole cells. By using a cell-permeable HDAC substrate, the activity of various lysine-specific protein deacetylases can be measured in intact cells in a simple and homogenous manner. The fluorescence of the deacetylated reaction product can be analyzed using a fluorometer with an excitation wavelength of 340-360 nm and an emission wavelength of 440-460 nm. An HDAC inhibitor, trichostatin A, is included for checking specificity of the HDAC reaction. This assay parallels Cayman’s HDAC Activity Assay Kit (Item No. 10011563), which uses a nuclear extract rather than whole cells for the assay. Together, both assays will help identify potential inhibitors/activators of HDAC enzymes.
