Tobacco Etch Virus nuclear-inclusion-a endopeptidase or TEV is highly sequence specific cysteine protease with N-terminal GST tag. It has a strict 7 amino acid cleavage recognition sequence of Glu-Asn-Leu-Tyr-Phe-Gln↓(Gly/Ser). The high specificity makes this protease excellent for the removal of affinity-tags from purified recombinant proteins. Purified TEV was expressed in E. coli and is shipped in 4°C packaging to preserve activity. TEV cleavage is best performed at room temperature in a pH range of 4 – 9.
