Ready-to-use lysates are especially prepared as positive controls for separation by SDS-PAGE and subsequent western blot analysis. Lysates are prepared in denaturing buffer WITHOUT dissociating agents (i.e. no 2-mercaptoethanol or dithiothreitol has been added). Heat lysate to 95° C for 5 minutes and rapidly cool. If dissociating conditions are desired, add reducing agent prior to heating. The recommended loading volume per lane is 10-20 µL depending on the size format of your gel.

A DNA sequence encoding the Human SLC39A10 (Q9ULF5)(Leu338-Phe831) was expressed with a polyhistidine tag at the C-terminus. This protein was prepared using our Nanodisc platform. Nanodisc is a versatile tool for studying membrane proteins. Using styrene-maleic acid (SMA) copolymer, membrane proteins can be extracted directly from prokaryotic and eukaryotic expression systems in the absence of detergents to preserve the protein structure and function better. Compared to membrane scaffold proteins (MSPs) nanodiscs, SMA nanodiscs also have the advantage of preserving proteins' nature by maintaining native lipids surrounded without introducing any heterologous proteins, which allows studies of protein structure and functions in a native-like environment.

The protein encoded by this gene is a coiled-coil-forming protein that associates with the SNARE (soluble N-ethylmaleimide-sensitive fusion protein attachment protein receptor) complex of proteins and the BLOC-1 (biogenesis of lysosome-related organelles) complex. Biochemical studies have identified additional binding partners. As part of the SNARE complex, it is required for vesicle docking and fusion and regulates neurotransmitter release. The BLOC-1 complex is required for the biogenesis of specialized organelles such as melanosomes and platelet dense granules. Mutations in gene products that form the BLOC-1 complex have been identified in mouse strains that are models of Hermansky-Pudlak syndrome. Alternative splicing results in multiple transcript variants.
