This MAb reacts with an extracellular domain (close to transmembrane) of CD56/NCAM. Three isoforms of neural cell adhesion molecule (NCAM) are produced by differential splicing of the RNA transcript from a single gene. The 135 kDa isoform is the basic molecule, which is glycosylated or sialylated to produce the mature species. Anti-CD56 recognizes two proteins of the neural cell adhesion molecule, the basic molecule expressed on most neuroectodermally derived tissues and neoplasms (e.g. retinoblastoma, medulloblastomas, astrocytomas, neuroblastomas, and small cell carcinomas). It is also expressed on some mesodermally derived tumors (rhabdomyosarcoma). Fibronectin is a soluble dimeric glycoprotein of 440 kDa, which is present in cells, extracellular matrix, and blood. This MAb reacts with the cellular as well as plasma form of fibronectin. Thyroglobulin is a 660 kDa dimeric pre-protein with mutiple glycosylation sites. It is produced by and processed within the thyroid gland to produce the hormone thyroxine and triiodothyronine. Prior to forming dimers, thyroglobulin monomers undergo conformational maturation in the endoplasmic reticulation. The vast majority of follicular carcinomas of the thyroid will give positive immunoreactivity for anti-thyroglobulin even though sometimes only focally. Poorly differentiated carcinomas of the thyroid are frequently anti-thyroglobulin negative. Adenocarcinomas of other-than-thyroid origin do not react with this antibody. This antibody is useful in identification of thyroid carcinoma of the papillary and follicular types. Presence of thyroglobulin in metastatic lesions establishes the thyroid origin of tumor.
