IL7 (interleukin 7) was first identified in 1988 in a bone marrow culture as a factor that facilitated murine B cell precursors growth. It is a cytokine produced by non-lymphoid cells in lymphoid organs. In humans it is also produced by keratinocytes peripheral blood dendritic cells follicular dendritic cells intestinal epithelial cells hepatic tissue endothelial cells smooth muscle cells and fibroblasts. IL7 gene is localized to human chromosome 8q12-q13. It is a glycoprotein with a four a-helix structure with a hydrophobic core. Recombinant human IL-7 is a 17.4 kDa protein containing 153 amino acid residues.

3-D Life is the most flexible system for biomimetic hydrogel design for 3D cell cultures. It uses two polymers that form a hydrogel by covalent crosslinking at ambient temperature. The chemical reaction is non-toxic to cells and occurs rapidly, preventing cell settlement before gel is formed. 3-D Life Dextran-HA Hydrogel Kit enables set up of hydrogels containing hyaluronic acid (hyaluronan). Thiol groups on crosslinker HyLink form stable thioether bonds with thiol-reactive groups on SG-Dextran, resulting in gel formation. Mixed at physiological pH 7.2 for optimal cell compatibility. Slow gelation allows for manipulation before onset of gel formation. Cell adhesion peptides (RGD, Cat. No. 09-P-001) can be attached to SG-Dextran to provide a cell-adhesive matrix. Dextran hydrogels can be dissolved by dextranase (Cat. No. D10-1), allowing easy cell recovery. One kit allows formation of 3.3 ml hydrogel of a crosslinking strength of 1.2 mmol/L. Insulin-Transferrin-Selenium-Ethanolamine (ITSE) is a basal media supplement and can be used to reduce the amount of fetal bovine serum (FBS) needed to culture cells. Insulin promotes the uptake of glucose and amino acids, transferrin is a non-toxic iron carrier, selenium is required for the function of glutathione peroxidase and other antioxidant enzymes, and ethanolamine is a phospholipid precursor. The components of the solution are manufactured animal-free and it is provided as a 100x sterile concentrate.
