3-Hydroxykynurenine (3-HK) is a metabolite in the kynurenine pathway of tryptophan catabolism, primarily involved in various physiological and pathological processes. It is a branch of the kynurenine monooxygenase (KMO) pathway and has been recognized for its dual role in the central nervous system. Structurally, 3-HK is an intermediate metabolite that undergoes autoxidation, which results in a range of products that may contribute to protein modification and cytotoxicity, especially in the context of oxidative stress. Although 3-HK has antioxidant and free radical scavenging properties, it is largely characterized as a toxic metabolite that can induce oxidative damage and cell death. The neuronal damage associated with 3-HK is mediated by free radicals rather than glutamate receptors. Altered levels of 3-HK have been observed in patients with various disorders, suggesting its potential involvement in neurodegenerative diseases.
