Rabbit Polyclonal Antibody

RNA demethylase ALKBH5 (UniProt Q6P6C2; also known as Alkylated DNA repair protein alkB homolog 5, Alpha-ketoglutarate-dependent dioxygenase alkB homolog 5) is encoded by the ALKBH5 (also known as ABH5, OFOXD1) gene (Gene ID 54890) in human. More than 100 types of postranscriptional RNA modifications are known, among which N6-methyladenosine (m6A) is the most common RNA modification. m6A modification mainly occurs in the consensus seqeunce motif RRm6ACH (R = A/G and H = A/C/U) located in stop codons, long internal exons as well as 3&prime UTR. RNA methyltransferases and demethylases are responsible for maintaining the level of cellular RNA m6A methylation. Two Fe2+ and 2-oxoglutarate (2OG) dependent AlkB dioxygenases family members, the fat mass and obesity-associated (FTO) enzyme and ALKBH5, catalyze the oxidative demethylation of abundant m6A residues. Both FTO and ALKBH5 are localized to nucleus and ALKBH5 gene knockdown is shown to affect cellular mRNA splicing in vitro and result in sterility phenotype in male mice in vivo.
