A suggested positive control for this product is HeLa whole cell lysate.Apoptosis, or programmed cell death, occurs during normal cellular differentiation and development of multicellular organisms. Cell death signals are transduced by death domain containing adaptor molecules and members of the caspase family of proteases. The human and mouse homologues to C. elegans death receptor 4 (CED-4) have recently been isolated and named Apaf-1. Apaf-1 binds to cytochrome C and caspase-9 (also known as Apaf-2 and Apaf-3, respectively), leading to activation of caspase-9. Activated caspase-9 activates caspase-3, leading to the proteolytic cleavage of a number of different proteins involved in the apoptotic cascade. Apaf-1 is ubiquitously expressed in human tissues and has also been shown to interact with caspase-4 and caspase-8.
