Inactivation of the BRCA1 gene, located at 17q21, is responsible for some forms of familial breast and ovarian cancer and sporadic ovarian cancer. BRCA1 encodes a protein of 1863 amino acids with an amino-terminal domain that exhibits similarity to the zinc-ring domain found in several regulatory and transcription factors. Mutations that disrupt this domain are some of the most frequently occurring ones linked to breast and ovarian cancer. Inhibition of BRCA1 expression leads to accelerated growth of mammary cells in vitro while loss of expression is observed during progression from benign disease to metastatic breast cancer suggesting that BRCA1 serves as a negative growth regulator. The ∽220kDa BRCA1 protein is expressed predominantly in the nucleus and appears to undergo cell cycle regulation with mRNA expression induced prior to DNA synthesis.
