Gangliosides are sialic acid-containing oligoglycosylceramides. The German scientist Ernst Klenk used ganglioside in 1942 to refer to lipids isolated from ganglions. Gangliosides are grouped based on the number of sialic acid residues per molecule, GM gangliosides contain one sialic acid residue, GD gangliosides contain two, GT contain three, GQ contain four sialic acid residues. Sialyltransferase-catalyzed α2-3 sialylation of the galactose of the trisaccharide Galβ1-4Glcβ1-1'Cer produces GM3, which can be further sialylated to produce GD3 with two sialic acid on the galactose. A de-N-acetylated form of ganglioside GD3 has been detected in human melanoma tumors. In addition, O-acetylation or lactonization of the sialic acid residue adds to the potential complexity. Gangliosides containing O-acetylated sialic acids, such as 9-OAc-GD3, occur in certain tumors and may protect cancer cells from apoptosis. GD3 is also known as CD60a. 9-O-acetylated GD3 is known as CDw60 or CD60b, while 7-O-acetylated GD3 is known as CD60c.
