NOS oxidizes a guanidine nitrogen of arginine releasing nitric oxide in the form of a free radical and citrulline. Nitric oxide thus generated acts as a mesenger in diverse functions including vasodilation neurotransmission, anti-tumor and anti-pathogenic activities. Inducible NOS (iNOS) or macrophage NOS (mNOS) is a calcium/calmodulin independent and is expressed in activated macrophages and stimulated glial cells.Heat-mediated antigen retrieval is recommended prior to staining, using a 10mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 10 min at room temperature. A suggested positive control is lung tissue.
