The 4E10 monoclonal antibody specifically binds to Junctional Adhesion Molecule-Like (JAML) which is also known as JAML1, AMICA, Amica1, Crea7, mCrea7, and Gm638. JAML is a type I transmembrane glycoprotein whose extracellular region contains two immunoglobulin domains. The 4E10 antibody binds to the membrane-proximal D2 Ig domain of JAML which is expressed on g ranulocytes, monocytes, memory T cells and γδ T cells. JAML serves as an endogenous ligand for epithelial and endothelial Coxsackie and Adenovirus Receptor (CAR). The JAML-CAR interaction supports leucocyte migration and the costimulation of TCR-triggered epithelial γδ T cells. The 4E10 antibody can induce costimulatory signals through JAML leading to increased activation, cytokine production and proliferation by epithelial γδ T cells.
