Description
MEF2D is a member of the MEF2 (Myocyte-specific Enhancer-binding Factor 2) transcription factor family. Members of this family (MEF2A, B, C, & D) contain several characteristic motifs that include a MEF2 domain and a MADS box. MEF2 proteins bind the DNA sequence, 5′- (C/T)T(A/T)(A/T)AAATA(A/G)-3′, and binding occurs primarily, although not exclusively, in muscle cells. MEF2D gene expression is ubiquitous and the mRNA is alternatively spliced. This results in multiple protein isoforms that heterodimerize with other family members. In addition to the MEF2 and MADS domains, MEF2D has a glutamine-/proline-rich region, amino acids 365-404, and a basic region, in the MADS domain near the N-terminus. The glutamine-/proline-rich region is involved in transactivation, while the basic region mediates sequence-specific DNA binding. MEF2D displays 75% similarity to hMEF2A and hMEF2C. Myogenin gene transcription requires MEF2D, indicating that MEF2D is involved in myogenesis and muscle cell differentiation. MEF2D has been reported to have a calculated molecular weight of 56kDa, but can be observed migrating in a range of 56-70kDa.
