The MLH1 gene is one of at least 4 genes encoding proteins involved in the repair of mismatch nucleotides following DNA replication or repair. Mutations in the MLH1 gene contribute to the development of sporadic colorectal carcinoma, while germline MLH1 mutations are responsible for ∽35% of inherited, non-polyposis colorectal carcinoma (HNPCC). MLH1 encodes a protein of 756 amino acids.
