PDI is an abundant component of the ER that catalyzes disulfide bond formation in the biosynthesis of various secretory and cell surface proteins. It is a multi-functional polypeptide identical to theβ-subunit of prolyl 4-hydroxylase and a cellular thyroid hormone-binding protein. It has two thioredoxin-like regions, each con-taining a Cys-Gly-His-Cys sequence that may represent two independently acting catalytic sites of PDI activity. The C-terminus of PDI has a KDEL sequence, which acts as retention signal for proteins of the ER lumen. PDI exists as a homodimer of∽107kDa. Prolyl-4-hydroxylase, anα2β2 tetramer, catalyzes the formation of 4-hydroxyproline in collagen by the hydroxylation of proline residues in peptide linkages. The PDI/β-subunit of prolyl-4-hydroxylase is required for its retention in the ER.
