Protein tyrosine phosphatases (PTPases) make up an expanding family of transmembrane and intracellular proteins, which function to remove phosphate groups from protein phosphoryl- tyrosine residues. The prototype phosphatase, PTPase 1B, was initially identified as an active intracellular phosphatase from human placenta made up of 321 amino acids and subsequently shown to derive from a larger precursor protein containing 435 amino acids. Using probes to the conserved PTPase domain a cDNA clone was isolated from a human peripheral T cell library encoding a protein with 65% identity to the placental PTPase 1B. The T cell protein differs in amino acid composition but is highly similar with respect to hydrophobicity with the 1B protein and is now known to be present in other cell types. While the function of the T cell PTPase protein is still unknown, recent experiments demonstrated that a truncated form overexpressed in BHK cells resulted in a greatly reduced growth rate of the cells. PTPases are involved in regulating processes where protein kinases and protein tyrosine phosphorylation are important. One intriguing hypothesis is that PTPases have the potential to function as anti-oncogenes.
