The T cell receptor or TCR is a plasma membrane protein complex responsible for recognizing antigens bound to major histocompatibility complex (MHC) molecules on antigen presenting cells. In 95% of T cells, the TCR is composed of a disulfide-linked alpha (α) and beta (β) dimer, while among 5% of T cells the TCR is composed of a gamma (γ) and a delta (δ) dimer. T cells are activated when their surface TCRs engage with antigen/MHC complex on antigen presenting cells. Each TCR alpha and beta chain contains a variable (V) and a constant (C) extracellular immunoglobulin-like domain, followed by a transmembrane region and a short cytoplasmic tail. The C domain lies proximal to the plasma membrane, while the V domain engages the antigen/MHC complex. Similar to immunoglobulin light chain and heavy chain, the TCR alpha and beta genes (Gene ID 21473 and 21577, respectively, in murine species) undergo VJ and VDJ recombination to generate TCR diversity for recognizing a wide range of antigens. the alpha and beta chains have very short cytoplamic tail and reply on the CD3 (cluster of differentiation 3) co-receptor for transducing signaling further intracellularly.
