TIP60 (accession no. Q92993) is a 60kDa protein that is a member of the MYST family, which is a distinct class of histone acetyl-transferases (HAT). TIP60 was initially identified as a protein that interacts with the HIV protein, Tat, and shares no homology with the HATs, CBP/p300, or PCAF. TIP60 was recently shown to modulate DNA repair and p53 function. TIP60 also directly acetylatesγ-H2AX and, together with the cofactor, TRRAP, binds to the chromatin surrounding sites of DNA double-stranded breaksin vivo. Furthermore, TIP60 associates with MYC and recruits it to the chromatin, which results in MYC-induced actetylation of histone H4. TIP60 directly associates with p53 and specifically acetylates it at K120, modulating the decision between cell cycle arrest and apoptosis. TIP60 acts as a transcription coactivator of p53 that enhances the activation of p21 and puma, leading to cell growth arrest and apoptosis. K120 actetylation is essential for p53-mediated puma activation and apoptosis, but is dispensable for both p53-mediated growth arrest, through p21 activation, and p53/MDM2 interaction. K120 is not acetylated by p300 and its actetylation by TIP60 does not affect the stability or DNA binding activity of p53.
